Building regions in PyTough

How can I build regions using pytough? I thought regions may be created by assigning rock properties to block centers, but it seems like that's not the way to build regions. Say, for example, I want to define 4 different regions in a sugar-cube type of mesh...how can it be done using pytough? Also, how can I assign boundary conditions and initial conditions to those specific regions?
